چکیده

Purpose ; Effects of topically applied 0.1% dernmethrsone on epithelirl and endothelial healing following alkali wound were examined. Mehtods ; 30 New Zealand white rabbits were divided in 3 groups. By using a 5.5 mm round filter paper, alkali wounds ( 1N NaOH, 60s ) were produced in one eye of each animal. Group A treated with 0.1% dexamethasone, 3 x/day for 3 weeks. Group B received 0.1% dexamethume, 3 x/day for the first week and then followed for 2 weeks without treatment. Group C served as control. Epithelial healing was determined by defect area measurement after fluorescein staining. Endothelial healing was measured at 1 and 3 weeks by morphometric determination following Alizarine red/trypane blue staining. Results ; Initial epithelial healing was retarded in dexamethasone treated group ( 0.57 f 0.06 mm’lh vs. 0.78 f 0.11 mm’/b, p-zU.01). However, group B showed better mtintanence of the resurfaced epithelium than group A or group C during the late healing. The endothelial defect area at 1 week was 14.1 f 1.31 mm’in treated eyes and completely resurfaced endotheiium was observed in group C. At 3 weeks, the endothelial defmt areas in group A and group C were 7.2 f 4.7 mm’ and 10.8 + 2.2 mm’, respectively. No endothelial defect was observed in group B. Conclusion ; Although the topical application of 0.1 % duamethnsone for the initial 1 week administration retarded the initial epithelial healing, it can delay the oecurance of the secondary endothelial breakdown process in comeal alkali wound healing.

Effect of Pseudomonas aeruginosa elastase, alkaline protease, and exotoxin A on corneal proteinases and proteins.

PURPOSE To determine the effects of exoproducts from the corneal pathogen Pseudomonas aeruginosa on corneal proteinases and proteins. METHODS Whole rabbit corneas were cultured in the presence or absence of broths conditioned with Pseudomonas aeruginosa, elastase, alkaline protease, and exotoxin A. Protein synthesis was assayed by adding 35S-methionine during the last 6 hours of culture. Case...

Elevated levels of matrix metalloproteinases in bronchoalveolar lavage fluid of emphysematous patients.

BACKGROUND Matrix degradation in emphysema has long been attributed to the action of neutrophil elastase (NE). More recently a role for other proteases, particularly the matrix metalloproteinases (MMPs), in the pathogenesis of this disease has been proposed. To date, however, the presence of MMPs in the lungs of patients with emphysema has not been demonstrated. METHODS Samples of bronchoalve...

Alpha-1-antitrypsin-human leukocyte elastase complexes in blood: quantification by an enzyme-linked differential antibody immunosorbent assay and comparison with alpha-2-plasmin inhibitor-plasmin complexes.

An enzyme-linked immunosorbent assay (ELISA) has been developed for the quantification of alpha 1-antitrypsin-human leukocyte elastase (alpha 1AT-E) complexes. In the ELISA, the alpha 1AT-E complex is bound to a surface by rabbit antileukocyte elastase antibody, and the inhibitor-proteinase complex is quantified by a second antibody, rabbit anti-alpha 1-antitrypsin F(ab')2, labeled with alkalin...

Fucoidan a sulfated polysaccharide from brown algae is a potent modulator of connective tissue proteolysis.

Fucoidans are sulfated fucosylated polymers from brown algae cell wall that exhibit some heparin/heparan sulfate properties. We previously demonstrated that these polysaccharides were able in vitro to stimulate dermal fibroblast proliferation and extracellular matrix deposition. Here, we investigated the action of a 16kDa fucoidan fraction on parameters involved in connective tissue breakdown. ...

Polymorphonuclear leukocyte activation and hemostasis in patients with essential thrombocythemia and polycythemia vera.

Thrombohemorrhagic complications are a major cause of morbidity and mortality in patients with essential thrombocythemia (ET) and polycythemia vera (PV). The pathogenesis of these complications is not completely clarified. Several studies have described abnormalities of red blood cells and platelets in these patients. However, no studies are available on changes in the polymorphonuclear leukocy...
